: Patients with type 2 diabetes mellitus (T2DM) are characterized by an acceleration of thrombopoiesis, an increase of platelet turnover, and a decrease of platelet survival time. These conditions affect the platelet count that manifest as an increase of the proportion of large and reactive platelets which are more thrombogenic. This study aimed to obtain the correlation between the platelet count and the platelet aggregation value in T2DM patients. This was an observational analytical study using the Pearson correlation test. Subjects were 30 T2DM outpatients at Endocrine-Metablolic Clinic of Prof. Dr. R.D. Kandou Hospital Manado. Blood samples of all patients were examined for platelet count and platelet aggregation value. The results showed that most of the subjects were ≥50 tahun (86.7%). Mean thrombocyte count was within normal level. Platelet aggregation values of the ADP 10 µm showed hypoaggregation, meanwhile of the ADP 5 µm showed normoaggregation The Pearson correlation test showed that there was no significant correlation between the platelet count and the platelet aggregation value, either using 10µm ADP (P = 0.22) or using 5µm ADP (P = 0.08). Conclusion: There was no significant correlation between the platelet count and the platelet aggregation values in patients with type-2 diabetes mellitus at Endocrine-Metabolic Clinic of Prof. Dr. R.D. Kandou Hospital Manado

Activity-dependent NPAS4 expression and the regulation of gene programs underlying plasticity in the central nervous system

The capability of the brain to change functionally in response to sensory experience is most active during early stages of development but it decreases later in life when major alterations of neuronal network structures no longer take place in response to experience. This view has been recently challenged by experimental strategies based on the enhancement of environmental stimulation levels, genetic manipulations, and pharmacological treatments, which all have demonstrated that the adult brain retains a degree of plasticity that allows for a rewiring of neuronal circuitries over the entire life course. A hot spot in the field of neuronal plasticity centres on gene programs that underlie plastic phenomena in adulthood. Here, I discuss the role of the recently discovered neuronal-specific and activity-dependent transcription factor NPAS4 as a critical mediator of plasticity in the nervous system. A better understanding of how modifications in the connectivity of neuronal networks occur may shed light on the treatment of pathological conditions such as brain damage or disease in adult life, some of which were once considered untreatable. Activity Dependent Branching Ratios in Stocks, Solar X-ray Flux, and the Bak-Tang-Wiesenfeld Sandpile Model

We define an activity dependent branching ratio that allows comparison of different time series XtX_{t}. The branching ratio bxb_x is defined as bx=E[ξx/x]b_x= E[\xi_x/x]. The random variable ξx\xi_x is the value of the next signal given that the previous one is equal to xx, so ξx={Xt+1Xt=x}\xi_x=\{X_{t+1}|X_t=x\}. If bx>1b_x>1, the process is on average supercritical when the signal is equal to xx, while if bx<1b_x<1, it is subcritical. For stock prices we find bx=1b_x=1 within statistical uncertainty, for all xx, consistent with an ``efficient market hypothesis''. For stock volumes, solar X-ray flux intensities, and the Bak-Tang-Wiesenfeld (BTW) sandpile model, bxb_x is supercritical for small values of activity and subcritical for the largest ones, indicating a tendency to return to a typical value. For stock volumes this tendency has an approximate power law behavior. For solar X-ray flux and the BTW model, there is a broad regime of activity where bx1b_x \simeq 1, which we interpret as an indicator of critical behavior. This is true despite different underlying probability distributions for XtX_t, and for ξx\xi_x. For the BTW model the distribution of ξx\xi_x is Gaussian, for xx sufficiently larger than one, and its variance grows linearly with xx. Hence, the activity in the BTW model obeys a central limit theorem when sampling over past histories. The broad region of activity where bxb_x is close to one disappears once bulk dissipation is introduced in the BTW model -- supporting our hypothesis that it is an indicator of criticality.Comment: 7 pages, 11 figure

Chk1 requirement for high global rates of replication fork progression during normal vertebrate S phase

Chk1 protein kinase maintains replication fork stability in metazoan cells in response to DNA damage and DNA replication inhibitors. Here, we have employed DNA fiber labeling to quantify, for the first time, the extent to which Chk1 maintains global replication fork rates during normal vertebrate S phase. We report that replication fork rates in Chk1¿/¿ chicken DT40 cells are on average half of those observed with wild-type cells. Similar results were observed if Chk1 was inhibited or depleted in wild-type DT40 cells or HeLa cells by incubation with Chk1 inhibitor or small interfering RNA. In addition, reduced rates of fork extension were observed with permeabilized Chk1¿/¿ cells in vitro. The requirement for Chk1 for high fork rates during normal S phase was not to suppress promiscuous homologous recombination at replication forks, because inhibition of Chk1 similarly slowed fork progression in XRCC3¿/¿ DT40 cells. Rather, we observed an increased number of replication fibers in Chk1¿/¿ cells in which the nascent strand is single-stranded, supporting the idea that slow global fork rates in unperturbed Chk1¿/¿ cells are associated with the accumulation of aberrant replication fork structure

Visual cortex plasticity: A complex interplay of genetic and environmental influences

The central nervous system architecture is highly dynamic and continuously modified by sensory experience through processes of neuronal plasticity. Plasticity is achieved by a complex interplay of environmental influences and physiological mechanisms that ultimately activate intracellular signal transduction pathways regulating gene expression. In addition to the remarkable variety of transcription factors and their combinatorial interaction at specific gene promoters, epigenetic mechanisms that regulate transcription have emerged as conserved processes by which the nervous system accomplishes the induction of plasticity. Experience-depenDent changes of DNA methylation patterns and histone posttranslational modifications are, in fact, recruited as targets of plasticity-associated signal transduction mechanisms. Here, we shall concentrate on structural and functional consequences of early sensory deprivation in the visual system and discuss how intracellular signal transduction pathways associated with experience regulate changes of chromatin structure and gene expression patterns that unDerlie these plastic phenomena. Recent experimental eviDence for mechanisms of cross-modal plasticity following congenital or acquired sensory deprivation both in human and animal models will be consiDered as well. We shall also review different experimental strategies that can be used to achieve the recovery of sensory functions after long-term deprivation in humans. © Copyright 2012 José Fernando Maya-Vetencourt and Nicola Origlia
